What causes Sotos syndrome and what are its cardinal features?
An NSD1 pathogenic variant or deletion causes an autosomal dominant disorder, usually de novo. Its cardinal features are childhood overgrowth, a characteristic facial appearance, and learning or developmental difficulties. Macrocephaly is common; adult height may be less distinctive than childhood growth.
